Er gilt als bedeutendster Bau der Manuelinik (Manuel I., 14951521), einer portugiesischen Variante der Spätgotik, die auch einige Elemente der Renaissance enthält. Unter anderem beherbergt es die Sarkophage von Fernando Pessoa, Vasco da Gama und Lus de Camµes und verschiedener portugiesischer Könige. Durch die weite Parkanlage vor dem 300 Meter langen Gebäude kommt die reichverzierte Kalksteinfassade voll zur Geltung. In den Seitenflügeln befinden sich das Marinemuseum und das Archäologische Museum. Das Kloster wurde von Manuel I. in Auftrag gegeben, kurz nachdem Vasco da Gama von seiner ersten Indienreise zurückgekehrt war. Der Bau wurde zu großen Teilen durch die nun sprudelnden Einnahmen des berseehandels finanziert. Im Jahre 1502 wurden die Arbeiten aufgenommen, die von zahlreichen Architekten und Baumeistern begleitet wurden, unter ihnen Diogo Boitaca (Planung und Ausführung) und Jo£o de Castilho (Logistik). Der Bau wurde im Jahre 1544 fertiggestellt. Während der Herrschaft König Johanns III. wurde das Kloster um den Chor erweitert.
